Reconstitution of lyophilized cultures:

Closed vials must be kept at temperatures between 5-25°C until processed. When growing, maintain the optimal conditions as given in the catalogue of cultures.

1.
Snap the scored top end of the vial.

2.
Score the vial with a file.

3.
Disinfect the surface of the vial.

4.
Snap the scored vial and remove the inner vial containing the freeze-dried material.

5.
Dissolve the dried material by adding few drops of broth and stir up.

6.
Inoculate the suspension into a liquid medium, the recommended agar medium can be inoculated simultaneously.

Most cultures grow within a few days, but some strains need longer time for cultivation. It is a good idea to keep the original vial with the dissolved dry matter in a fridge for another possible inoculation.
